Abstract:

Larval red drum (10 to 14 days post-hatch) were assessed for visual function using behavioral and morphological assays. Optomotor response was determined using a rotating drum apparatus. Behavioral tests were video recorded and analyzed for a maximum rotational speed of larvae exhibited during the assay. Larvae were subsequently preserved and sectioned histologically to determine alterations in retinal layers due to oil exposure, with sections photographed and examined using ImageJ software (V1.47). Purpose:

To determine impacts of oil exposure on visual function in larval red drum using behavioral and morphological assays.

Data Parameters and Units:

Dataset contains 2 folders (Histology_images and R4.x258.0000050_Videos) and one excel file (Eye histology measurements). Folder "Histology_images": The image filenames are defined as Number_dph_dose_RDreplicateNumber_histologicalCassette_SlideNumber.jpg where dph = days post-hatch, RD = Red Drum, dose refers to either control experiment or 1% oil from a surface (OFS). If there is a “2” after the “SlideNumber”, it means that there were two images taken for this individual fish, as one image shows both left and right eyes, and the other just the left eye. Folder "R4.x258.0000050_Videos": The video filenames are defined as Number_dph_dose_DD_Mon_YY_FishNumber_exposure_RotationSetting.mp4 where dph = days post-hatch, dose refers to either control experiment or 1% oil from a surface (OFS). The rotation settings include forward spin direction of optomotor behavioral assay (F), reverse spin direction of optomotor behavioral assay (R), not following alternating stripe in behavioral assay (N), following alternating stripe in behavioral assay (T), following in the same direction of an alternating stripe in behavioral assay (same direction). Eye histology measurements.xlxs: slide number, days post-hatching (dph), dose (1 for 1% OFS or 0 for control), replicate number, whole retina (micrometers), lens (micrometers), ganglion (micrometers), inner plexiform (micrometers), inner nuclear (micrometers), outer plexiform (micrometers), outer nuclear (micrometers), photoreceptor layer (micrometers), and pigmented epithelium (micrometers) were analyzed in each retina.

Methods:

Images of each fish were taken on an Axio imager A1 Zeiss compound microscope and the diameter and area of each layer were analyzed using ImageJ software (Version 1.47). Red drum embryos were collected from broodstock tanks at the Texas Parks and Wildlife-CAA Marine Development Center in Corpus Christi. High-energy water accommodated fractions (HEWAFs) were generated by mixing 1 g of slick crude oil with 1 L of saltwater by blending on low for 20 s with a commercial blender. Following mixing, the HEWAF was poured into a separatory funnel where it settled for a duration of 1 h. Approximately 2,500 embryos were loaded into a 9 L pickle jar that contained either control saltwater or dilutions of HEWAF, where they remained until 24 hours post fertilization. Red drum were then transferred to 164 L conical mesocosms containing clean seawater for the duration of the experiment.
